  • The white part of this is fresh undercoat going onto old oil based gloss paint. The crazy part is, that cream colour used to be bright white. 
This is what happens to white oil based paint which is deprived of UV light. This is why we use water based paint on woodwork as out first choice, it dries quicker, is low odour and most importantly it stays white :) #oilbased #waterbased #acrylic #hmg #hmgpaints #uvlight #oldoil

  • Genomic DNA isolated from S. pombe

We first open (lyse) the cells using glass beads, a combination of buffers are added to the tubes which are spun at a high speed (centrifuged) to generate three distinct layers in the tubes. The DNA in the top layer is carefully removed and precipitated into propanol. This solution is centrifuged again to get a small transparent pellet which is resuspensed into solution again. Once the concentration and quality of the DNA is checked we can then use it in downstream applications such as PCR.

The concentration and quality is first determined using spectrophotometry (nanodrop) where a 260/280 absorbance ratio between 1.8~2.0 indicates pure DNA. We can also determine the concentration of DNA using this method. Our concentrations averaged 100 ng/ul and the absorbance ratios were 2. So far so good but the quality of the DNA must still be checked.

Good quality DNA is checked by agarose gel electrophoresis and then imaged by fluorescence under UV light. Quality DNA is seen by a clean band near the top of the gel (depending on genome size).
